Abstract

Diabetes Mellitus is a disease caused by decreased pancreatic function to produce insulin or insensitivity of insulin receptors. This research aims to analyse the bioactive compounds in spoon leaves as antidiabetics and the mechanism of action in silico. Bioactive compounds were collected using Dr. Duke's Phytochemical and Ethnobotanical Databases. Compound unification was done through PubChem. Way2Drug PASS Online was used to analyse biological activities related to antidiabetic. SEA and Swiss Target Prediction databases to determine the target protein. Molecular docking was performed using Pyrx application and visualized with Biovia Discovery Studio. A total of 8 spoon leaf bioactive compounds targets alpha glucosidase, alpha amylase, and GLP-1 proteins that regulate the occurrence of diabetes with a Pa>0.5 value. Based on the docking results luteolin, plantagoside, nepetin and scutellarin are alpha glucosidase inhibitors with binding affinity values of -7.5, -8.7, -8.2 and -8.3 kcal/mol. Catalpol and neochlorgenic acid are alpha amylase inhibitors with binding affinity of -7.1 and -7, 3 kcal/mol. Aucubin and fumaric acid are GLP-1 acceptors with binding affinity values of -7.3 and -4.6 kcal/mol. Visualization of docking that nepetin and neochlorgenic acid compounds, have similar interactions and amino acid residues with control drugs so that they can be used as antidiabetic drug candidates.
